Reduced insulin binding to liver plasma membrane and adipocyte of vacor rodenticide poisoned rats

Insulin binding to liver plasma membranes and adipocytes of 5 normal and 7 Vacor-poisoned rats has bene investigated to find extra-pancreatic factor responsible for Vacor-induced diabetes mellitus. Wistar rats were poisoned with low dose (5 mg/kg body weight) of Vacor to avoid an early death and yet to allow the development of glucose intolerance. Liver plasma membranes and adipocytes obtained from Vacor-poisoned rats at the 4th day after Vacor administration had a reduced insulin binding capacity as compared to normal ones. Scatchard analysis showed that the reduction of 125I-insulin binding in the Vacor-poisoned rats was due to a decrease in the number of insulin receptors in the membranes and adipocytes. These results suggest that Vacor-induced diabetes mellitus is at least in part due to the reduction in the number of insulin receptors in the peripheral tissues of Vacor-poisoned subjects.
