The amounts of nutrients recommended by the nrc support sustained ethanol metabolism in rats in vivo

The amounts of nutrients recommended by the NRC abate the effects of a toxic alcohol dose administered to rats. The objective of this study was to determine whether the inadequate amounts of nutrients ingested with the 36% liquid ethanol diet result in a decreased rate of ethanol metabolism relative to the rate of ethanol metabolism suppported by the amounts of nutrients recommended by the NRC and ingested with the 26% liquid ethanol diet. Rats were fed either the 26% or the 36% liquid ethanol diet for ten days. The rats were continued on their respective diets and their diet intake measured hourly for the next 42 hours. The hourly diet intake was used as the data input into a blood flow limited physiological pharmacokinetic model of ethanol metabolism in the rat. The simulation studies showed that rats ingesting inadequate nutrients with the 36% liquid ethanol diet were unable to metabolize ethanol as fast as rats ingesting adequate nutrients with the 26% liquid ethanol diet. The blood alcohol concentration was always higher in rats ingesting the 36% liquid ethanol diet as was the time the blood ethanol concentration exceeded 20 mM. Hence, the area under the blood ethanol concentration versus time curve was significantly greater in rats ingesting the 36% liquid ethanol diet. In addition, there was a major temporal difference in alcohol ingestion which resulted in an additional peak in blood ethanol concentration which occurred 20-25 h after the start of the study. The decrease in the rate of ethanol metabolism when nutrients are deficient may be accounted for either by a decrease in the activity of the low Km alcohol dehydrogenase, a decrease in activity of the high Km alcohol metabolizing enzymes (alcohol dehydrogenase, alcohol P-450 oxygenase or catalase) or by a combination of the two effects.
