The impact of dna ploidy and proliferative index on survival of breast cancer patients

Flow cytometric DNA analysis was performed on 135 samples of breast cancer. The DNA ploidy was found to be diploid in 31 cases (23%) and aneuploid in 104 cases (77%). The DNA index ranged from 0.70 to 2.07. The determination of proliferative index (PI: the fraction of tumor cells in the S + G2M phase of the cell cycle) was possible in 94 cases. The pI ranged from 1.4% to 62.9% and it was greater than 30% in 21 cases. DNA ploidy of tumors did not correlate with any of conventional prognostic variables but lymphatic invasion within the tumor. DNA ploidy did not correlate either with the survival of patients. PI of tumor cells appeared to correlate with stage of the disease, lymphatic invasion within the tumor, nodal involvement, and microscopic infiltration of the tumor into the adjacent tissues. Higher PI (greater than 30%) correlated closely with poorer survival of patients and more frequent relapse of the disease. Multivariate analysis revealed that PI of tumor cells, microscopic infiltration of the tumor into the adjacent tissues and tumor size correlated with the survival duration of patients after surgery, in order of decreasing significance. Lymph node involvement, histologic type and PI of tumor cells, correlated with the disease-free survival duration of patients after surgery, in order of decreasing significance.
