Aspergillus niger CFTRI 6, a soil isolate, produced merely 5.5 g citric acid per kg wheat bran in solid state fermentation system involving basal wheat bran medium against that of 16.3 g .cntdot. L-1 in submerged fermentation process. The %-conversion works out to be 6.8 in solid state fermentation, thereby indicating that the titres and the yield are poor from an industrial point of view. The culture required additional nitrogen, carbon and phosphate for enhanced production of citric acid in solid state fermentation system. Incorporation of 3% methanol in the medium was found to improve citric acid production significantly. The standardization of these parameters resulted in a 13.1 times increase of citric acid production as compared to that in basal wheat bran medium, though the %-conversion was 32.8. The data indicate a high potential of Aspergillus niger CFTRI 6 for citric acid production in solid state fermentation system.
