Lipoxygenase is a key enzyme to cause beany flavour in soybean seeds. This study was undertaken to determine the seed lipoxygenase activity of 23 soybean genotypes. Three substrates for lipoxygenase, which were Arachidonic acid, Linoleic acid and cis-Linoleic acid, were used to measure varietal difference in that activity. Identifying soybean genotype with low or high seed lipoxygenase activity was independent of the kind of substrates. Kwangkyo belongs to the group which showed the low lipoxygenase activity, whereas Danyeobkong, Jangyeobkong and Muhankong had high activity in recommended varieties. An improved SDS - polyacrylamide gel electrophoresis was used to detect the presence of lipoxygenase isozymes. And also, new techniques using the carotene bleaching activity of lipoxygenase were applied to determine the presence or absence of the lipoxygenase isozymes. Carotene bleaching test could clearly indentify the existence of lipoxygenase isozymes. Both SDS-PAGE and carotene bleaching test were available for observing lipoxygenase isozymes. The latter was more rapid and simple, whereas the former was more accurate.
