Breeding of feedback inhibition resistant mutants of Eremothecium ashbyii overproducing riboflavin

E. ashbyii is an important filamentous fungus overproducing riboflavin. The synthetic medium in which E. ashbyii grows excellent was designed on the basis of the investigation of its nutrient requirement. Three mutants, U-95-1, U-95-2 and U-96-3 obtained from original strain of E. ashbyii treated with U. V by selected on the synthetic medium containing 8-AG of analogue of guanine (50 mu-g/ml), were selected to determine their productivity and stability in the procedure of continual inoculation by flask fermentation. Results showed that the productivity of mutants U-95-1 and U-95-2 increased 15.5% and 9.8% respectively compared with start strain and also showed good genetic stability after five to ten generations. A way to improve productivity and stability of E. ashbyi was provided.
