Plastid DNA of the moss Physcomitrella patens (Hedw.) B.S.G. was digested with BglII/BglII/EcoRI, ClaI, HindIII, EcoRI, or EcoRI/PstI, respectively. The resulting fragments were cloned onto pUC 13 and integrated into a physical map by hybridization, restriction analysis and partial sequencing. For this purpose an efficient method for the detection of ptDNA in recombinant plasmids utilizing the dot-blot-technique and the ECL-system (Amersham) was developed. Up to now, the identified and integrated ptDNA-fragments comprise 51% of the circular ptDNA molecule. Restriction analysis of these clones with six different endonucleases resulted in an improved partial physical map of the plastid DNA. Furthermore, partial sequence analysis revealed a high homology of plastome encoded genes between the moss Physcomitrella patens and the liverwort Marchantia polymorphia.
