Effect of actinomycin D on systemic acquired resistance of tobacco plants to tobacco mosaic virus

Effect of actinomycin D (AMD), 10 mu-g/ml, on systemic acquired resistance (SAR) of hypersensitive tobacco plants Nicotiana tabacum L. (cv. Xanthi nc) to tobacco mosaic virus was studied. AMD had been injected intercellularly 3 hours before challenge inoculation of the upper leaves or was added to incubation medium of protoplasts. AMD did not affect the number of lesions, but the lesion size and TMV accumulation in leaves of both control and repeatedly infected plants, and protoplasts isolated from these leaves were increased in the presence of AMD. SAR was almost completely abolished by AMD, when plants were challenged 3 days after inducing inoculation. AMD did not affect the SAR 7 days after the inoculation. It is supposed that resistant factors formed in initially infected leaves and transported in uninoculated ones induce syntheses of virus inhibitors.
