In vitro culture of soybean immature embryo VII. Histological studies organogenesis and embryogenesis from soybean immature embryo cultures

Immature embryo axes of soybean cultivar Kaoshing No. 10 were cultured in 1/2 MSB medium with 0.6 mg/l BA for inducing organgenesis of shoots and calli. Immuture cotyledon were also cultured in MSB medium with an addition of 2.5 mg/l 2, 4-D for inducing embryogenesis of somatic embryo. During the cultural processes, paraffin section method was used to study the histology of the organs and somatic embryos, which they produced. The results are giving in the following. 1. The multiple buds produced by organgenesis were the results of a series of differentiation events: primarily, the meristem cell divided into parenchyma, followed by the formation of primordium, and shoot. Finally, the vascular bundle was developed. 2. The rough-shaped callus induced from explants might form tracheal cells with a nest-like or strand-like structure and then differentiated into roots. 3. The embryogenesis started from the division of the cells with the ability to form somatic embryos, then and followed by four developed stages, they were globular, heart-shaped, torpedo-shaped and dicotyledon structure, finally, became mature embryos. 4. The explant could form abnormal somatic embryo with monocotyledon. This kind of somatic embryo were not capable of sprouting.
