Abstract
PURPOSE A specific, accurate and reproducible RP-HPLC method for simultaneous determination of methaqualone and its 2'-hydroxymethyl metabolite in biological material was developed. METHODS A Waters HPLC instrument with mu-Bondapak C-18 Radial column was used. A mobile phase was composed of CH-3OH:NaH-2PO-4 buffer solution(60:40) at the flow rate of 1.6 ml/min, 490 uv detector was operated at 235 nm. RESULTS: Serum concentrations of methaqualone ringing of 1 apprx 35 mg/L was in good linear relationship. The average recovery was 100.15% (RSD lt 3.9%). The minimal detectable concentration was 15 mu-g/L. 2'-hydroxy methyl-metabolite was gained by TLC and was proved by MS. It was determined according to the linear concentration of methaqualone. CONCLUSIONS: This method is proved to be useful in simultaneous determining methaqualone and 2'- hydroxy methyl-metabolite in biological material by a volunteer and a toxic body.
