A study on the transformation of the tobacco plant (Nicotiana tabacum Br21, N. tabacum NC82) using Ti-plasmid vector pEND4K containing Zea mays transposable element(Ac) was carried out to obtain a mutant tobacco cell line and to use gene tagging system. Transformed Escherichia coli and Agrobacterium tumefaciens were confirmed by the resistant ability to the appropriate antibodies and the size of DNA restriction fragments. Transformed tobacco cells were selected in the selection media (MS) containing 0.5 mg/l of 2, 4-D, 2 mg/l of kinetin, 100 mg/l of kanamycin and 150 mg/l of carbenicillin.
