A dual role for the kinase-like domain of the tyrosine kinase Tyk2 in interferon-a signaling

The function of the kinase-like (KL) domain of Tyk2 in interferon (IFN)a/Gb signaling was investigated. Point mutations in the KL domain that are responsible for loss-of-function phenotypes were identified from a genetic screen of randomly point-mutated Tyk2 cDNAs, and the IFNa signaling pathway in cell lines that expressed these mutants were analyzed. The results showed that an intact KL domain is crucial for the inherent catalytic activity of Tyk2 and for the establishment of a high-affinity interferon-a receptor complex.
