A genome scan and linkage analysis was conducted using a large European kindred without superoxide dismutase 1 mutation and with classic autosomal dominant adult-onset amyotrophic lateral sclerosis (ALS). A novel locus was identified. At recombination fraction 0.0, the maximum LOD score is 4.5 for polymorphism D18S39. A 7.5 cM, 8 Mb region of chromosome 18q21 that was flanked by markers D18S846 and D18S1109 was identified as a novel familial ALS locus.
