The activation of the bacteriophage Mu mom promoter (Pmom) was studied. The phage-encoded C protein, a site-specific DNA binding protein, is known to be involved in activating transcription from Pmom, as well as from 3 other late operons. In vitro transcription analyses revealed that the activation of transcription from Pmom did not require specific interactions between C and the C-terminals of either the a or s70 subunit of Escherichia coli RNA polymerase. This is contrary to previous results for the other Mu-encoded activator Mor and is surprising considering the high sequence identity between C and Mor.
