Determination of the mechanism of orotidine 5'-monophosphate decarboxylase by isotope effects

Kinetic 15N isotope effects for the enzymatic decarboxylation of orotidine 5'-monophosphate (OMP) are reported. An effect of 1.0036 was obtained for OMP, compared with 0.9955 for decarboxylation of picolinic acid at 190[degree]C and 1.0053 for N-methyl picolinic acid at 120[degree]C. The driving force for the OMP decarboxylation appears to be ground-state destabilization due to charge repulsion between the carboxyl of the substrate and Asp-91.
