In vitro delivery kinetics of glyceryl trinitrate and isosorbide dinitrate from the transdermal therapeutic systems Nitropercuten and Sorbopercuten

The in vitro delivery kinetics of glyceryl trinitrate (GTN) and isosorbide dinitrate (ISDN) from the matrix transdermal therapeutic systems (TTS) Nitropercuten and Sorbopercuten through the cadaveric human skin epidermis and the model polymer membrane Carbosil was investigated. The TTS Nitropercuten and Sorbopercuten were demonstrated to ensure high drug delivery rates, 75% of GTN and ISDN of their baseline matrix content being delivered from TTS by the zero-order kinetics. In accordance with the first Fick diffusion law, the release rate of the two agents from TTS was proportional to their concentrations in the diffusion matrix. The correlation coefficients between the release rate of GTN and ISDN in vitro via the polymeric membranes and human skin were 12.9 and 37.0 for GTN and ISDN, respectively. The in vitro delivery kinetics of GTN via the membrane Carbosil from the TTS Nitropercuten, Nitroderm, Nitro-Dur, and Nitrodisc was determined.
