The interactions between tumor necrosis factor (TNF) and 3 modifiers of its activity in natural killer cells were examined. Listeriosis in mice with the severe combined immunodeficiency (SCID) mutation is a model of interferon c-dependent macrophage activation by natural killer cells in the development of natural immunity. The effect of interleukin- (IL) 2, IL-10, IL-12 and TNF-a on the production of interferon- (IFN) c in response to heat killed Listeria monocytogenes by SCID spleen cells was assessed. Data indicate that macrophage production of TNF-a and IL-12 stimulated the release of INF-Gg by the natural killer cells, and IL-10 inhibited it at the level of the macrophage and the killer cell. It was concluded that IL-12 and TNF-a are costimulators of INF-c production. .
