The aggregation state of Ure2p in Saccharomyces cerevisiae strains carrying the [URE3] prion was investigated. Ure2p is a regulator of nitrogen catabolism in yeast; loss of the [URE3] genetic element results in loss of nitrogen catabolite repression. Fusions of Ure2 with green fluorescent protein were found to form intracellular aggregates specifically in [URE3] strains. The [URE3] prion was cured by the overexpression of such fusion proteins and certain fragments of Ure2p. These results suggest a means to a treatment of other prion diseases.
