Cucumber mosaic virus and tobacco mosaic virus proliferation in transgenic chili peppers and its protoplast

Transgenic chili pepper plant expressing CMV and TMV coat protein gene were infected with CMV and TMV to comparing the characteristic of CP-MR in transgenic chili pepper and virus content in inoculated plants. The results showed that the transgenic chili pepper exhibited high level of coat protein-mediated resistance to both CMV and TMV. The development of systemic symptom was 7apprx15 days delayed, percentages of plants showing symptoms and severity of disease symptom in inoculated plants were strongly reduced, and virus accumulat and spread were obviously repressed in inoculated and newly-developed leaves in CP(+) plants challenged with CMV and TMV. The protoplasts from transgenic chili pepper expressing CMV-CP and TMV-CP were infected with CMV virion. The results showed that protoplasts from transgenic chili pepper can also inhibit the virus proliferation. The protoplasts were infected with 40mug/mL virus concentration, 48 hours later, the amount of virus in control plant protoplasts was 4.2 times than that in CP(+) plant protoplasts. This results demonstrated that the resistance characteristic of transgenic chili pepper was to reduce virus proliferation in plants.
