Effect of inhibiting tumor angiogenesis on cell proliferation and apoptosis in mice with bladder cancer

Objective: Our aim was to study the effect of inhibiting tumor angiogenesis on cell proliferation and apoptosis in mice with bladder cancer. Methods: N-butyl-N-(4-hydroxybutyl) nitrosinmine (BBN) was used to develop mouse bladder cancer. TNP-470 30 mg (15 mg/kg) was injected intraperitoneally twice every week. The microvascular density (MVD) of tumor, the proliferative index (PI), and the apoptotic index (AI) were investigated. Results: The MVD in the treated group by TNP-470 was 15.31+-3.66, significantly lower than that in control group (P<0.01). The PI in the control group was from 5.2% to 41.1% (average 21.7%), and that in the treated group was from 9.6% to 21.6% (average 15.0%). There was evident difference between the two groups (P<0.05). The AI in the control group was from 1.0% to 4.3% (average 2.3%). The AI in the treated group was from 1.1% to 20.7% (average 6.81%). There was significant deference between the two groups (P<0.01). Conclusion: TNP-470 can inhibit the cell proliferation in mice bladder cancer, and accelerate tumor cell apoptosis or necrosis.
