Ethanol inhibits gene expression of nerve growth factor in primary cultures of rat astrocytes

In order to elucidate the mechanism of ethanol impairment on nerve system, primary cultures of astrocytes from the cerebral cortex of neonatal rats were exposed to ethanol at concentrations of 0.5-5.0 g cntdot L-1. The levels of nerve growth factor(NGF) mRNA were analyzed and quantified by reverse transcription polymerase chain reaction (RT-PCR) and Northern blot methods. The results showed that the levels of NGF mRNA were decreased significantly by exposure to the ethanol at the concentration as low as 0.5 g cntdot L-1 for 24 h. The highest concentration of ethanol at 5.0 g cntdot L-1 produced 50% and 58% reduction of NGF expression after 48 and 72 h incubation compared to untreated cultures. Our results indicate that ethanol inhibits NGF synthesis of astrocytes in vitro. The reduction of NGF may lead to the disruption of neuronal-glia interaction and underlie, in part, the neuropathology associated with fetal ethanol exposure.
