Haploid induction in apple: First homozygous scab and mildew resistant lines of the apple cultivar 'Remo'

Haploid induction in apple: First homozygous scab and mildew resistant lines of the apple cultivar 'Remo'

Gartenbauwissenschaft 64(3): 134-137

The application of a developed protocol for anther culture and induction experiments of in situ parthenogenesis followed by embryo culture resulted in the production of 2 lines of the apple cultivar 'Remo' which were characterized as diploid, homozygous and resistant to scab and mildew. Ploidy level determination was done by flowcytometry and the homozygosity of the shoots was analysed by isoelectric focusing and polyacrylamide gel electrophoresis using 3 isozymes. After rooting, the plants were transferred to the greenhouse and the experimental garden. Following the first winter period, scab and mildew tests were carried out on potted plants. For further characterization of resistance, habit and growth, fruit quality and yield, the plants have been grafted and transferred into the selection orchard.
