Irradiation conditions for induction of nitrite reducing activity of horse heart cytochrome c

Denatured horse heart cytochrome (Cyt) c by gamma-irradiation showed high reducing activity of nitrite (NO2-) to ammonia (NH4+), in the presence of sodium dithionite and methylviologen. Effect of O2 on irradiation of the Cyt c and the detailed irradiation conditions for nitrite reducing activity of 0.03 mM Cyt c have been investigated. The Cyt c in an aqueous solution (0.03 mM) was treated by Co-60 gamma-irradiation at doses of 0.25-5.0 kGy, dose rates of 0.15-5.0 kGy/h under N2 or O2 atmosphere. The Cyt c babbled with O2 during irradiation showed much higher activity than that with N2. The excellent irradiation conditions for increasing the activity of 0.03 mM Cyt c were total dose of 1.0 kGy under O2 atmosphere. The activity of irradiated Cyt c by these conditions was increased approximately 5 times of unirradiated one.
