DNA alkylation properties of dehydromonocrotaline: sequence selective N7 guanine alkylation and heat and alkali-resistant multiple crosslinks

Pyrrolizidine alkaloids are a group of plant toxins that have been responsible for several episodes of fatal human poisoning and large-scale losses of livestock. In this work, the DNA alkylation properties of activated monocrotaline (a pyrrolizidine alkaloid) were examined using gel electrophoresis and electron microscopy. The results showed that dehydromonocrotalione reacts sequence selectively with DNA in the major groove.
