Glucoamylase production by Aspergillus spp. during solid state fermentation using wheat bran, yam peel and banana stalk as substrates

Aspergillus spp. produced significant level of glucoamylase using inexpensive agricultural wastes in solid state fermentation(SSF). Maximum enzyme yield of 490 U/ml was obtained on wheat bran after 3(rd) day at 40 degrees C by A. ochraceus (MTCC 1877). Aspergillus sp. (lab isolate) produced 410 U/ml of glucoamylase on yam peel after 4(th) day of incubation. On wheat bran amended medium, A. ochraceus showed specific activity 2.6 U/mg protein with purification factor of 4.14. The optimum pH for glucoamylase production under SSF was found to be 4.0 for A. ochraceus (MTCC 1877) and 6.0 for Aspergillus sp. (lab isolate). At 40 degrees C the glucoamylase production was maximum for the two species of Aspergillus. Among the substrates tried, wheat bran and banana fruit stalk favoured the maximum production of glucoamylase by A. ochraceus while Aspergillus sp. (lab isolate) preferred yam peel.
