Three species of mosquitoes were collected from Guangdong province, including Culex fuscanus, Cx. bitaeniorhynchus and Cx. pipiens quinquefasciatus. The mosquitoes' ribosomal DNAs in the internal transcribed spacer region 2 (ITS2) were sequenced. To explore the sequence divergences a neighbor-joining tree was established compared with other 12 mosquito species ITS2 sequences in GenBank. The homogeneity of ITS2 is 65.27% between Cx. fuscanus and Cx . tigripe, 75.87% between Cx. bitaeniorhynchus and Cx. pseudovishnui and 97.13% between Cx. pipiens quinquefasciatus and Cx. pipiens pollens . These results show that subgenus Lutzia is closely related with subgenus Culex. It is also certified the larval stages of the two subgenera mosquitoes bear genetic similarity. The subgenus Melanoconion, Neoculex and Lutzia are closely related with Aedes. The results support the traditional taxonomy based on morphology, but diversity presented more commonly in the subgenus Culex.
