Inoculation techniques for evaluating resistance to Pseudomonas solanacearum

The efficacy of various inoculation procedures in evaluating resistance to bacterial wilt or Granville wilt (P. solanacearum) in various crops was detd. under greenhouse conditions. Inoculations with virulent cultures of P. solanacearum either by puncturing the stem through a drop of bacterial suspension placed at a leaf axil or by pouring a bacterial suspension over wounded secondary roots were equally effective on young succulent seedlings of susceptible hosts (Marglobe tomato, Variety 402 tobacco or Irish Cobbler potato). In resistant tomato (F5 and F6 breeding lines), tobacco (Oxford 26, Dixie Bright 27, 101 and 102), and peanut (Schwarz 21) plants, disease indices obtained by stem-inoculations were higher than those obtained by root-inoculations. However, best differentiation between resistant and susceptible vars. was made on the basis of root-inoculations. Susceptibility to bacterial wilt decreased in resistant tomato plants as age of plants increased from 4 to 8 wks. Decreasing the inoculum concn. resulted in a decrease in disease severity in young resistant tomato plants but not in seedlings of a susceptible var.
