In rabbits fed with cholesterol some of the endothelial cells of the aorta contain droplets of lipid material in their cytoplasm. Most of these cells, and all of those most heavily laden, overlie or adjoin the aortic plaques, but small groups of such cells are present in areas that are otherwise apparently normal. Macrophages laden with lipid material are seen on both sides of the endothelium covering the plaques, and some laden macrophages can be demonstrated apparently passing through the endothelium. These observations clarify certain disputed points about the cellular distribution of lipid in the aorta of rabbits after cholesterol feeding but they do not reveal the nature of the initial lesion.
