The Japanese method of counting the number of chloroplasts in the guard cells of the stomata of the leaf is the quickest way of distinguishing between diploid, triploid and polyploid sugar-beet. The present paper describes improved techniques for making chloroplasts more easily visible. These include introducing a wetting agent to the cuticle and, by using a solution of I2 in KI, identifying the chloroplasts by means of their starch grains instead of darkening them with AgNO3.
