Various methods were tested. Of the indirect methods in which protein nitrogen is first determined and later converted into protein content, the approved Kjeldahl method proved accurate and reliable. The analysis of total and residual nitrogen by Nessler's reagent was not recommended, because of the dependence of the reaction on time and on pH. Of the direct methods, the biuret test was simple, time-saving, and sufficiently accurate.
