Hypersensitivity to Ascaris antigens. II. The skin-sensitizing properties of 7 S gamma 2 antibody from sensitized guinea pigs as tested in guinea pigs

Hypersensitivity to Ascaris antigens. II. The skin-sensitizing properties of 7 S gamma 2 antibody from sensitized guinea pigs as tested in guinea pigs

J. Immun, 99: 2, 347-356

guineapigs were immunized by weekly intramuscular injections of a crude pig Ascaris extract in complete Freund's adjuvant and bled 7 days later. Sera showing 7S gamma 1 and gamma 2 precipitin arcs by immunoelectrophoresis were pooled. Anti-ascaris antibodies were obtained from 3 separate pools as follows: by precipitation with ammonium sulfate at 33% saturation and subsequent electrophoretic separation on Geon blocks; j by decomplementation, ammonium sulfate precipitation and subsequent specific precipitation with, and dissociation from, a thiolated Ascaris extract; and by precipitation with ammonium sulfate only. Aliquots from all the preparations were absorbed with a rabbit anti-guinea-pig 7S gamma A serum and antigenically pure guineapig 7S gamma 2 anti-ascaris antibodies as well as unabsorbed controls were tested by passive cutaneous anaphylaxis (PGA) in guineapigs. The results indicated that guineapig 7S gamma 2 anti-ascaris antibodies were able to elicit local ana-phylactic reactions of the skin. Although the reaction appeared later (11/2 to 4 hours after challenge) than in the classical PCA, the possibility of a reserved passive Arthus-type reaction was ruled out.
