Half an udder, isolated from a lactating ewe, was perfused for 7 h with a substrate mixture containing methionine (28.6 micro c), and milk was collected every hour. The specific activity of casein increased linearly during the 1st 5 h of perfusion to reach a max. value after 6-7 h. Of the 35S incorporated into casein 99 % was in methionine residues while cystine residues had a weak specific activity.
