Leghemoglobin from alfalfa (Medicago sativa L. Vernal) was purified and fractionated into two major (LbII and LbIII) and three minor (LbI, LbIV and LbV) electrophoretically distinct components. Translation of poly(A)-containing mRNA from alfalfa nodules in vitro gave a full complement of the components and suggested that they were not derivatives of each other or artifacts produced during fractionation. Spectral analyses of the reduced pyridine hemochromogen showed protoheme IX to be the heme moiety of all five components. The components cross-reacted immunologically but had different amino acid compositions and were coded for by different messages. The two major components, LbII and LbIII, had very similar spectral characteristics in the oxidized or reduced states and as nicotinate complexes but there were differences in the peptide maps of their partial tryptic digests. Antibody prepared against the purified components cross-reacted strongly with nodule extracts of pea, clover and milk vetch but not with those of soybean, lupine, jackbean and black locust.
