Modifications in the direct determination of urea N by diacetyl monoxime have yielded very sensitive automated and non-automated procedures requiring low acid strength. A 2.0 ml/min. saline diffusate of blood (AutoAnalyzer) is combined with 2. 5 ml/min. of 2. 6 [image] sulfuric acid, 0.4 [image] phosphoric acid, 0.3 [image] ferric chloride and 1.6 ml/ min. 0. 25 percent(w/v) diacetyl monoxime and 0.05 percent (w/v) thiosemicarbazide. After heating to 95[degree]C and cooling, absorbance is measured at 543 mu in a 10 mm. flow cell. Statistical analysis (t test) indicates no significant difference at the 95 percent probability level between the automated method and a manual urease-Nesslerization technique. The standard deviation of the diacetyl method is 1.1 mg urea N/ 100 ml with a standard error of the estimate of 1.8 mg/ 100 ml. The sensitivity of the proposed modification may be increased by altering reagent strengths, however, in its present form, it has reduced previous acid requirement almost 10 fold with no loss in precision.
