Auxin production by the rice leaf blight fungus Helminthosporium oryzae and other species of Helminthosporium

Auxin synthesizing potentials of 7 species of Helminthosporium, H. oryzae, H. gibberosporium, H. tetramura, H. turcicium, H. nodulosum and H. setarige, were determined by growing them in tryptophan-Czapek's medium for 20 days in dark, extracting the auxin and estimating by spectrophotometric and chromatographic methods; all the species produced IAA in the medium, indicating that tryptophan was the auxin precursor. H. oryzae produced proteolytic enzyme in a casein-Czapek's medium and with higher proteolytic activity more auxin resulted. Tryptophan, the auxin precursor, was abundantly present in the casein medium. Accumulation of IAA in casein medium is attributed in part to its release from IAA -protein complex by the fungal proteolytic enzymes and to the conversion of tryptophan to IAA. Rice plants infected with H. oryzae exhibited higher auxin levels. The possible accumulation of IAA due to break down of the cell wall proteins by proteolytic enzymes during pathogenesis is suggested.
