The assay consists essentially of the vaccination by oral route of a group of mice, another like group serving as control. All animals are challenged by the oral route with a standardized living suspension of typhoid bacillus after a certain time, and bled from the heart in due time for bacterial counts. The difference between mean log counts in both groups gives a protective index, since bacteriemia is inversely related to resistance.
