Intraperitoneal injection of W2531 at 200 mg/kg into young female Wistar rats caused a significant (P. 0.001) inhibition of acetate-1-Cl4 incorporation into serum (76%) and liver (71%) cholesterol at 2 hours. After 4 and 18 hours the inhibition was less. No marked change was observed in either serum or liver cholesterol concentration in this short study. When W2531 was fed to normal adult male rats at 250 mg% or 400 mg% in a chow diet for 2 weeks, it signficantly inhibited acetate-l-Cl4 Incorporation into liver (55-65%) and serum (67-70%) cholesterol. At 400 mg% in the diet, W253I lowered serum cholesterol 35%, and in both dose levels lowered liver cholesterol 14-20%. Histological examination of the livers from the high dose rats revealed hypertrophy of the liver but no discernible pathological damage.
