Several methods of determining protein in blood serum were examined. Among the indirect procedures, which 1st determine the protein nitrogen and from it calculate the protein concentration, the Kjeldahl method proved to be exact and reliable if a greatly dissociated acid (sulfurlc acid) is used before distillation. Two percent boric acid is unsuitable. Determination of the nitrogen total and remainder with Nessler's reagens is not recommended because of the time and pH dependence of the reaction. Of the direct methods, the Biuret reaction is simple and time saving and sufficiently precise. It is the preferred method for the clinical laboratory.
