Persistent infection of HeLa and Detroit 6 cells by herpes simplex virus occurred only in the presence of specific antibodies from human serum. In the absence of these antibodies, e.g. after replacing human serum by horse serum, the virus began to multiply rapidly and the cells degenerated. After removal of the antibodies, the level of intracellular virus increases 32- to 6300-fold. The amount of intracellular virus in the presence of specific antibodies was only 0.015 -3.1% of the amount in cultures containing no antibodies. In persistently infected cultures, 1 median tissue culture infective dose of herpes simplex virus was produced by 60-100 cells. If hyperimmune rabbit serum was added to persistently infected cell cultures, the virus multiplied and the cells succumbed to the action of the virus. Cultures with antibodies added yielded the same amounts of intracellular virus whether incubated at 23[degree] or 36[degree] C. On removing the antibodies the virus multiplied at 36[degree] C, but at 23[degree] C production of the virus was inhibited. During the whole period of cultivation of persistently infected cells, the presence of virus manifested itself by isolated plaques with a morphology typical of herpetic infection.
