The DNA content, measured by Feulgen cytophotometry, of diploid ectodermal and entodermal cells is practically identical. The small nucleus of the binuclear spermatid in the male gonad of the polyp, as well as the small nucleus of the binuclear parasitic cell (the earliest known Polypodium stage in the sterlet egg), contain 2 times less DNA somatic cell nuclei, and are thus haploid. Nuclei of blastomeres of the cleaving embryoare also haploid; thus, no fertilization occurs in Polypodium, and diploidy seems to be restored at later stages of embryogenesis. The large nucleus of the spermatid in the male gonad of the polyp begins polyploidization just after meiosis and contains the DNA quantity corresponding to ploidy levels of 2 to 6 n. The large nucleus of the binuclear cell, which gives rise to the "capsule " around the parasite in the host egg, continues polyploidization attaining ploidy degrees of about 400 n. Nuclei of the nurse cells of the Polypodium ovary are also polyploid (8 to 32 n). Female sexual cells which leave the gonad of the polyp have diploid nuclei, thus corresponding to oogonia.
