Experiments have been performed which demonstrated that there are great variation in mitotic rate and in the rate of DNA synthesis of the epidermal cells of the back skin of hairless mice after a single application of 0.005 ml methylcholanthrene in benzene and that these changes roughly parallel each other. Mitotic rate and the rate of DNA synthesis are most rapid 24 hours after the application, when mitotic count and the number of DNA synthesizing cells are relatively low, and mitotic rate is only slightly above the "normal" rate on the 11th day, when mitotic count is markedly increased. Thus, mitotic rate and the rate of DNA synthesis cannot be correctly calculated from mitotic count and from the number of DNA synthesizing cells unless mitotic duration and DNA synthesizing time are constant and not influenced by the experimental treatment.
