Exaggerated delayed-type hypersensitivity to simple chemical allergens in the guinea-pig

Intradermal administration of small amounts of mycobacterla in paraffin oil markedly enhances the degree of sensitivity when simple chemical allergens are Injected. The mycobacterla may be injected prior to or within a few hr. after the intradermal injection. The method leads to satisfactory sensitizaticm even of a stock of guinea-pigs (Sewell Wright's Family XIII) that are rather refractory to sensitizatlon by allergenlc chemicals. Subsequent skin tests at weekly intervals markedly increases the sensitivity until a plateau is reached. This seems to be of an anamnestic response in a system of purely delayed-type hypersensitlvlty. Not only plcryl chloride and dinltrochloro-benzene behave in this way, but sensitizatlon to picric acid is markedly enhanced. Sensitlzatton to quinine and (to lesser degree) formaldehyde was attained. The method with proper variations, may allow superior testing in the guinea-pig of materials to be screened for potential allergenicity in the human being. Delayed sensitivity to ovalbumin developed under conditions in which a simple injection only gave traces of sensitivity.
