Methods of determining the contact activation index of blood clotting

The intimaloses the property of an unwettable surface which promotes the adherence of thrombocytes and leads to the development of thrombosis in lesions of the vascular wall. The zeta potential, which normally varies around 0 between the surface of the endothelium and the circulating blood, changes. The variations of the zeta potential can be determined on the basis of determining the degree of wettability since the zeta potential varies around 0 in undamaged blood vessels and in unwettable glass test tubes. The ratio of the clotting time in siliconized test tubes to that in ordinary glass was proposed as an index of wettability, reflecting the state of the contact factor. This index is called the contact activation index of the blood. The possibility of replacing siliconized test tubes by more accessible unwettable materials was investigated. The clotting time of healthy examinees and patients with obliterating endarteritis and various hematological diseases were studied simultaneously in ordinary siliconized and polyethylene test tubes. The clotting time of the blood of the healthy examinees in siliconized test tubes if 22. 7[plus or minus]4. 26 min. and in polyethylene ones, 27. 2[plus or minus]4. 4 min. The clotting time in the polyethylene test tubes was usually somewhat higher than in siliconized test tubes both for healthy persons and patients with various blood diseases. This indicates that polyethylene test tubes can be used in place of siliconized ones to determine clotting time under conditions of an appreciable decrease of contact activity.
