Corn and spinach were grown in nutrient solutions from which Fe was excluded for 2-3 weeks. After the 2nd leaf had appeared with signs of chlorosis, the upper leaves were homogenized and divided into 4 fractions by centrifugation. Determinations of RNA, DNA, Fe, and Mn were run on these fractions which consisted chiefly of: (1) nuclei, (2) nuclei + chloroplasts, (3) chloroplasts with nuclear debris, and (4) mitochondria with chloroplast debris. When Fe was excluded from the nutrient medium, the Fe content was considerably lower in nuclei and mitochondria and the RNA content was higher in all of these fractions. There were no appreciable changes in the DNA content. Changes in the Mn content were negligible in cellular structures in corn, but in spinach the exclusion of Fe from the nutrient medium resulted in a pronounced increase in the Mn content of the chloroplast and mitochondria fractions. The authors conclude that a direct relationship exists between the content of Mn and. RNA and an inverse relationship between Fe and RNA.
