Polarographic maxima were used to study the formation of complexes between DNA and proteins. Measurements were made on a series of tubes containing 1 ml crystalline proteinase from Bacillus subtilis (20 [mu]g/ml in 0.6 [image] NaCl) and 0.05-1 ml DNA from herring roe (50 [mu]g/ml) and from thyroid. Tubes were made up to 2 ml with 0.006 M NaCl after which 6 ml of maximum-forming reagents (Biokhimiya 26:575, 1961) were added. The process of complex formation in the absorption curves was represented by segments which were not a function of the concentration of one of the components in the complex (if the concentration of the other component is constant). Experiments with DNA depolymerization showed that in neutral medium and at pH 9 formation of a complex between DNA and subtilysin occurred in a 1:2 ratio. Highly polymerized thyroid DNA formed a complex in a 1:1400 ratio in a neutral medium (at complete saturation). Complexes of highly polymerized DNA and subtilysin are apparently formed by adsorption. It is possible that the inner surface of DNA's double helix also participates in complex formation. Polarographic maxima can be used to determine the degree of depolymerization of nucleic acids. Highly polymerized nucleic acids are able to suppress the maxima more than depolymerized DNA.
