Sub-cellular localization of 5-hydroxytryptamine and substance X in molluscan ganglia

Density gradient fractions of Mercenaria ganglia homogenate with high relative specific activities of 5-hydroxytryptamine (5-HT) and also containing some Substance X (a potent cardio-excitor) were examined under the electron microscope. An attempt was made to correlate the occurrence of the different types of small particles observed in different fractions with the relative specific activities of 5 -HT and Substance X, in order to determine the types of particles with which the substances are normally associated. Results indicated that Substance X is associated with "uniformly granulated vesicles" of 1000-3000 A dia. 5-HT is probably not associated with electron dense-cored particles (1000-2000 A dia.). The precise sub-cellular localization of 5-HT in molluscan nervous tissue is briefly discussed in the light of observations made.
