The activity of microsomes from regenerating rat liver in amino acid incorporating systems

Microsomal suspensions were prepared in parallel from normal and regenerating rat livers and were tested in systems containing standard amounts of normal cell fluid as the source of activating enzymes. The activity for amino acid incorporation into the proteins was determined with C14-L-leucine as the labeled component. To compare the incorporation rate between the two microsomal suspensions their contents of total nitrogen, protein, RNA and glucose-6-phosphatase were determined. On the basis of equal protein content the activities of the suspensions for leucine incorporation increased after a lag period of about 10 - 15 hours after partial hepatectomy. Full microsomal activation was normally reached after about 30 hours. Simultaneously with the rise there was an increase of RNA content in the microsomal fractions while the activity of glucose-6-phosphatase decreased. The change may indicate modified proportions between the granular and lamellar constituents of the endoplasmic reticulum, from which the microsomal material emanates. The increased microsomal activities during the regeneration period may be conditioned by a higher average concentration of nucleoprotein granules connected with the reticular membranes.
