The effect of nalorphine on morphine and codeine analgesia and lethality and the interaction of morphine and codeine

In rats, the LD50 dose of codeine (I) was found to be 140 mgAg intraperitoneally and 540 mgAg subcutaneouly. The LD50 of morphine (n) was 235 mg/kg intraperitoneally and 265 mg/kg subcutaneouly. The dose-response curve was much steeper for (I) than for (II). Nalorphine (III) 100 mg/kg increased the toxicity of (I), but decreased the toxicity of (II). Simultaneous administration of (I) and (n) potentiated the analgesic and lethal effects of both. The analgesic effect, measured by the hot plate method, of (I) was clearly reduced by (HI); about 0.6-07. mg/kg halved the effect The antagonistic effect of (HI) on (II) was smaller and much less pronounced.
