The effect of Na and K on the stimulation of the adenosine triphos-phatase activity and the inhibition of the p32-(ATP) adenosine triphos-phate exchange reaction of fresh liver mitochondria by triamcinolone and 2,4-dinitrophenol was studied. 2,4-Dinitrophenol stimulated the adenosine triphosphatase reaction and inhibited the P32-ATP exchange reaction equally well in Na and K media. Triamcinolone produced a marked stimulation of adenosinetriphosphatase in K medium, but not in Na medium; the inhibition of the P32-ATP exchange reaction was in the range of 90% in K medium and 50% in Na medium. Triamcinolone, but not 2,4-dinitrophenol, stimulated adenosinetriphosphatase activity at 0.35 M or higher concentrations of KC1. At 0.35 M KC1, the triamcinolone-stimulated adenosinetriphosphatase activity was unaffected by cyanide. The diminished P32-ATP exchange reaction of fresh mitochondria in 0.35 M KC1 was inhibited by triamcinolone, 2,4-dinitrophenol, and cyanide. Triamcinolone induces a rapid and drastic swelling of mitochondria in 0.125 M KCL but if K is substituted by Na, only a slight swelling effect is observed.
