The histochemical demonstration of histamine in mast cells

The successful histochemical demonstration of mast cell histamine depended on adequate fixation. When rat tissues containing mast cells were frozen-dried and fixed in formaldehyde vapor, several histochemical reactions applied to these tissues indicated the presence of histamine in the mast cells. Diazonium coupling with diazotized p-bromoaniline, fluorescence and reducing properties were all consistent with the presence of intracellular histamine which has reacted with formaldehyde. Reactions performed on aqueous formaldehyde fixed tissues were weakly positive or negative.
